Optimized matrix substance is intended for preparation of matrix

solution for mass spectrometry by simple addition of organic solvent.

- All samples and inoculated products have to be considered as potentially infectious and obvious safety valid regulations must be respected while handling.

- Before use, check if the packaging is intact. Do not use damaged kit. Leftovers of expired or devaluated reagent liquidate according to valid regulations. Packaging materials are recyclable.

 

Diagnostic kits are delivered in multilayer packets on base of aluminium and organic polymers. Part of each packet is silica gel desiccant. Store kits at temperature from +2 to +25°C. Expirationdate is placed on each packaging. Store opened product at dark and dry place, with temperature under +25°C. Product can be stored one

week in such conditions (or to date of expiration, in case when it passes earlier).

 

Preparation of inoculum

 

DIRECT INOCULATION METHOD

- Dissolve content of microtube in 0,25 ml of organic solvent and homogenize till total dissolution of crystals

- Spread biological material in thin homogeneous layer on MALDI plate

- Cover spreaded sample with 1 μl of matrix.

- Let dry at laboratory temperature

- Analyse in device for mass spectrometry MALDI

Tip: Upper mentioned method, can be modified: spread one colony at one position on MALDI plate and cover it with 1μl of 70 % solution of formic acid after drying. When dried, cover sample with 1 μl of prepared substance

- Spread 1μl of axtracted material in homogenous layerat one positon of MALDI plate

- Dry at laboratory temperature

- Cover sample with 1μl of matrix substance

- Dry at laboratory temperature

- Analyse in device for mass spectrometry MALDI

 

Work with material as with potentially infectious agens. Liquidate leavings according to internal procedures and legislative of your country.
